Recent Advances in Generalized Pustular Psoriasis (GPP)

Generalized Pustular Psoriasis (GPP) is an enigmatic, chronic skin inflammatory disease, developing independently or in association with preexisting plaque psoriasis. Since the IL36RN mutation was revealed in some GPP cases in 2011, understanding of molecular patho-mechanisms of GPP has begun rapidly advancing in this decade. Namely, mutations/variants of IL36RN or CARD14 or AP1S3 have been found to be the cause of some GPP. Moreover, animal models of the disease have been developed recently and been analyzed to reveal immunological aspects of GPP. Meanwhile, many new medicines and therapies have also been developed and therapeutic management of this disorder is also progressing.

Although outstanding progress has been made in the GPP research field, molecular patho-mechanisms of this disease are still not completely understood. Treatment is also still challenging in some cases of GPP. In this Research Topic of Frontiers in Medicine, we introduce current knowledge of GPP both clinically and scientifically, and aim to clarify the prevailing problems in the research field and how they should be addressed.

Accordingly, this Research Topic welcomes the following themes:

•              Definition, clinical features, differential diagnoses (ACH, SPD, PPP, AGEP, etc), and comorbidities of GPP

•              Phenotypes of GPP and related diseases (autoinflammatory keratinization diseases)

•              Epidemiology of GPP

•              Patho-mechanism of GPP from an immunological standpoint

•              Patho-mechanism of GPP from a genetic standpoint

•              Treatments of GPP (Biologics)

•              Treatments of GPP (Non-biologics)

•              Animal model of GPP Il36rn knock out mic

•              Animal model of AIKD (Card14 gain of function mutation)
